I am currently comparing the Gl 450 2008 Honda pilot 2009 and the Toyota highlander 2008, I really don't know which to go for, I am more inclined towards the pilot as I used the 06 model before but I would like to give the Benz a try, Is there anything I should be afraid of as regards the Benz?

I am also interested in fuel economy, efficiency, maintenance and other factors?

The GL doesn't belong on the list if you are looking for fuel economy.
The ml350 can be considered.
Get the four cylinder highlander. It has the best fuel economy of the lot.

MDX>Highlander>2007 RX350

The only car that'd give you decent fuel economy is the four cylinder Highlander. Small engine in an SUV must be frustrating, though.
